  • The Solid Reactor is a catalyst for copper-catalyzed azide-alkyne chemistry (CuAAC) which does not need to be dissolved during the reaction unlike other Cu catalyst (copperbromid, coppersulfat) for catalyzing it. For this reason, it has the advantage that the concentration of dissolved copper ions is very low and therefore it is an ideal choice for performing click reactions on copper sensitive material like RNA. Additionally the Solid Reactor can easily be removed after the reaction is finished by simply removing it and does not need additional steps as soluble catalysts. For ideal working conditions a ligand as THPTA (BCMI-006) is needed to be added to the reaction mixture.

    • Which reagents to avoid in copper catalyzed click reactions?

      Some biochemical buffer components can interfere with the copper-catalyzed click reactions (CuAAC). For example, TE and TAE buffer contain EDTA which chelates the copper ions and thus decreases reaction kinetic and yield. Moreover, free thiols, as dithiothreitol (DTT) and mercapto ethanol, can deteriorate or completely prevent click reactions.

    • What kind of labels can be clicked?

      As it is the nature of click reactions, various groups can be attached to nucleic acids, not constricted to dyes. The only requirement is the use of high quality reagents containing clickable groups.
